I am planning a cross country road trip and I want to visit car culture related places along the lines of the Pistenklause in Nurburg and the Seneca Lodge at Wakins Glen.
Provide some examples around the USA?
So far I have Classic Car Club Manhattan, the Corvette Museum, Tail of the Dragon to name a few.

Is America not car culture? The second most expensive purchase most people ever make is a car. Does a car not sometimes hold a family member status even to those not blessed with the love of cars? Do we not travel daily in our vehicles and do they not remind us of home or carry memories that last our lifetime? All of America is car culture.
Anyway enough of that bull shit. I suggest you consider Detroit. Henry Ford Museum, Auburn, Cord Duesy museum in Indiana. Both are relatively close and any car culture trip in the USA needs to include Detroit.

Chatterbox Drive In Diner, but you need to go during the summer on a Saturday evening for their cruise night. Hundreds of muscle cars, hot rods, the occasional exotic and everything in between. If you’ve never been on a Saturday night, you literally have no idea how awesome the event is.

The Larz Anderson Museum just outside of Boston is pretty neat if you can go there on a day where there’s a lawn event.

And if you’re in this area, take the detour north to check out the Gilmore Museum, especially on one of the weekends where they’re having a car show (nearly every one during the summer). My personal favorites are Deutsche Marques (all german) and the Red Barn Spectacular (1200+ cars).
That being said, even with nothing going on, the museum itself is absolutely worth the trip.

Katie’s Cars and Coffee in Great Falls, VA and THE Cars and Coffee that was in Irvine, CA but has relocated to places like the Orange County Fairgrounds in Costa Mesa . It’s only fair that you should attend one of the most major C&C events and then the one that started it all...even if it’s moved, the car culture that made the original happen will be there.

If you pass through Nebraska, be sure and check out the Speedy Bill Smith Museum of American Speed. If you love racecourse and America, it’s a must-see. Also, if you can get an invitation (it’s private), check out the Kuck collection.

If you decide to visit Indianapolis Motor Speedway and the Auburn, Cord, Duesenberg Museum, let me know. I live less that 20 minutes for IMS and about 1.5 from Auburn, IN.
